The present work summarizes the isolation and characterization of human satellite and intermediate DNAs. Satellite DNAs were obtained from native high molecular weight DNA by Ag+-Cs2SO4 centrifugation. Two components of intermediate DNA, called $\alpha$ and $\beta$ were obtained by preparative CsCl centrifugation of total human DNA renatured to a "Cot" of 20. The $\alpha$ component was obtained only from low molecular weight DNA, while the $\beta$ component was obtained also from high molecular weight DNA. Hypotheses on the function of repeated DNAs are discussed.
