The citoplasmic "petite" mutation which arises in yeast spontaneously with a frequency of 1/102-1/103 is induced with an extremely high efficiency by acridines and by a number of other agents including ethidium bromide, U.V., fluoro-uracil, etc. The base composition of mitochondrial DNA in mutants is profoundly altered and the study of the effect of these agents on DNA replication could provide an explanation of the molecular mechanism of the mutation. The effect of some mutagenic and related non-mutagenic dyes on DNA replication in vitro has been tested in the presence of Escherichia coli and yeast mitochondrial DNA polymerase. The results indicate a preferential replication, in the presence of mutagenic dyes, of A+T rich regions of DNA.
