In connection with their researches on the mechanism of action of theskin-photosensitizing furocoumarins, the AA. have recently pointed out the possibility of a photoreaction «in vitro» between the same furocoumarins and DNA, after irradiation at 3.655 Å. Such a photoreaction is now confirmed and more extensively studied using a labeled furocoumarin, i.e. bergapten - O14CH3. The formation has been ascertained of a chemical bond between the furocoumarin and DNA, after irradiation at 3.655 Å. Moreover it has been found that the furocoumarins photoreact also with the pyrimidine bases and their nucleosides and nucleotides but they do not with the corresponding purine derivatives. Therefore, the AA. postulate that the pyrimidine bases may be the reactive sites also in DNA. With RNA and thermally denaturated DNA the furocoumarins photoreact in a much lesser extent. Since this photoreaction is specific for the skin-photosensitizing furocoumarins, it appears likely that it could explain the mechanism of the till now observed biological photosensitizing effects of furocoumarins, as the skin-erithema, the DNA- viruses inactivation and the lethal and mutagenic action on bacteria and on mammalian cells adapted to in vitro growth.
